🔬 How it works: a small interfering RNA that switches off PCSK9 production inside the liver cell at the messenger RNA level. LDL receptors survive longer, the liver clears more cholesterol from the blood. A GalNAc conjugate targets the molecule almost exclusively to the liver. 📊 Evidence: LDL cholesterol fell about 50% versus placebo on top of statins (ORION-10 and ORION-11, NEJM 2020) and about 40% in familial hypercholesterolaemia (ORION-9); the effect held over years of follow-up (ORION-8).
